Sulfobutylether-β-Cyclodextrin (SBE-β-CD, sodium sulfobutylether β-cyclodextrin) is an anionic highly water-soluble β-cyclodextrin derivative. It has been officially included in the Chinese Pharmacopoeia (2025 Edition, Volume IV) as a pharmaceutical excipient. Featuring a hydrophilic exterior and hydrophobic inner cavity, SBE-β-CD forms non-covalent inclusion complexes with drug molecules to enhance solubility and stability of poorly soluble drugs, reduce hemolysis and mucosal irritation, making it suitable for parenteral formulations.
Its primary application field is pharmaceutical formulation. It is widely adopted in injectable products including voriconazole, posaconazole and ziprasidone injection. It can also be applied in ophthalmic, nasal and oral preparations. It exhibits strong inclusion affinity toward cationic nitrogen-containing drugs, improving bioavailability and masking unpleasant odors of active pharmaceutical ingredients. It can stabilize proteins by inhibiting aggregation in biologic formulations, and deliver hydrophobic compounds in in-vitro cell research.
Reported research directions in published literature cover its application in novel drug delivery systems such as microneedles, in-situ gels and nanoformulations; molecular modeling for inclusion complex prediction to accelerate formulation screening; investigation of its potential for toxin binding and antidote use; optimization of substitution degree control and quality standard system to support the development of biologics and innovative small-molecule drugs.
